近期，中國醫(yī)學科學院皮膚病研究所姚煦教授團隊、復旦大學附屬華山醫(yī)院李巍教授團隊及香港城市大學董立新教授團隊合作在 Cell Host & Microbe（IF=20.6）上發(fā)表題為“Multi-omics Signatures Reveal Genomic and Functional Heterogeneity of Cutibacterium acnes in Normal and Diseased Skin."（多組學特征揭示正常和患病皮膚中痤瘡丙酸桿菌的基因組和功能異質(zhì)性）的研究，在該項目中，研究團隊從1234個分離基因組中證明了C. acnes的個體和生態(tài)位異質(zhì)性。皮膚疾?。裾詈宛畀彛┖蜕眢w部位決定了C. acnes的基因組差異，這種差異源于水平基因轉(zhuǎn)移和選擇壓力。與表皮葡萄球菌（Staphylococcus epidermidis）相比，C. acnes具有獨-特的代謝功能，抗生素耐藥基因和毒力因子較少，基因組更穩(wěn)定。綜合基因組、轉(zhuǎn)錄組和代謝組分析在菌株水平揭示了C. acnes的功能特征。與轉(zhuǎn)錄組特征一致，C. acnes在富含皮脂的環(huán)境中對角質(zhì)形成細胞產(chǎn)生毒性和促炎作用。L-肌肽（L-carnosine）是一種抗氧化應(yīng)激代謝物，在濕疹的代謝組中上調(diào)并減輕皮膚炎癥?？偟膩碚f，該研究揭示了基因和微環(huán)境對C. acnes功能的共同影響。利用從健康皮膚、濕疹（AD）和痤瘡患者皮膚分離的痤瘡丙酸桿菌（Cutibacterium acnes, C. acnes）菌株進行了全基因組測序、轉(zhuǎn)錄組測序、代謝組測序以及功能研究，研究團隊首-次從單菌株層面揭示了C. acnes的基因和功能異質(zhì)性。通過深入地了解宿主和皮膚微生物群之間的復雜關(guān)系，有望能找到增進皮膚健康和改善皮膚病管理的靶點，也為將來探索C. acnes的臨床應(yīng)用奠定了基礎(chǔ)。
