瘦素(Leptin)是由ob基因編碼、在脂肪組織中生成的一種脂肪代謝調(diào)控產(chǎn)物，在代謝和調(diào)控體重等方面發(fā)揮重要作用。它通過下丘腦中的瘦素受體發(fā)出信號，降低食欲，增加能量消耗。在外周組織中，瘦素能拮抗胰島素信號傳導，增加脂肪酸氧化，減少胰島β細胞的胰島素分泌，并促進生育。它在脂肪細胞中的表達可以被其他脂肪因子如IL-6（刺激）和TNF-α（抑制）以旁分泌方式調(diào)節(jié)。瘦素缺乏的小鼠表現(xiàn)出嚴重的胰島素抵抗、肥胖和生育能力下降，所有這些都可以通過外源性瘦素給藥而逆轉(zhuǎn)。還有研究表明，瘦素與心血管疾病有關，因為它介導了許多促炎和促動脈粥樣硬化作用，如內(nèi)皮素-1（ET-1）表達增加、活性氧（ROS）積累、單核細胞趨化蛋白-1（MCP-1）分泌以及交感血管張力和血壓增加。
